The research investigates the mechanisms behind Lewy body disorders, focusing on alpha-synuclein and its role in extracellular vesicle (EV) dynamics. By employing patch-clamp electrophysiology and real-time molecular imaging, the study explores the release kinetics of EVs in hippocampal neurons. A novel pH-sensitive molecular tag was developed to analyze the fusion of multivesicular bodies with the plasma membrane, while mass spectrometry was used to identify molecular interactions related to Lewy body pathology. This work aims to enhance understanding of disease progression and potential therapeutic targets.
